Expressions of TRPV1 and TRPA1 in the dorsal root ganglion in the rat model of orchialgia

Objective: To explore the molecular mechanism of ochaialgia by studying the expression of TRPV1 and TRPA1 in rats.
Material and Method: The models of orchialgia were established in male SD rats by the injection of 2% acetic acid into the center of testes．Behavioral test were conducted to record the number of pain response and withdraw latency. The expressions of TRPV1 and TRPA1 in T13-L1 DRG were determined by RT-qPCR, Western blot and Immunofluorescence.
Result: Compared with the control rats, the orchialgia models showed a significant increase in number of pain response and a decreased withdraw latency (P<0.05). Both TRPV1 and TRPA1 were expressed in the membrane of neurons in DRG. Compared with control, the expression of both ion channels were increased significantly (P<0.05).
Conclusion: The up-regulated expression of TRPV1 and TRPA1 in DRG may associate with the allodynia and hyperalgesia in the rat model of orchialgia.
